Abstract

This study aimed to observe the morphological changes in neuronal cells in primary cultures of mouse brain (Mus musculus) during incubation. Primary cell culture is an efficient method for studying the dynamics of neuronal development in vitro while maintaining the original biological characteristics. The study utilized DMEM culture media enriched with FBS and Penicillin-Streptomycin, with neuronal morphology observed on days 3, 6, 8, 9, and 10 using an inverted microscope.

The results showed significant morphological changes. On day 3, neuronal cells appeared round without neurite branching, indicating an early adaptation phase. On day 6, neurons began to exhibit early differentiation by forming short neurite branches. Further differentiation occurred on day 8, marked by the development of intercellular connections through elongated neurite branches. By days 9 and 10, neurons displayed mature morphology with complex network structures, extensive neurite branching, and stable intercellular connectivity.

This study concludes that primary cultures of mouse brain cells can represent the stages of neuronal proliferation and differentiation progressively, achieving mature morphology. These findings provide a relevant in vitro model for neurobiological studies, including neurodegeneration mechanisms and cell-based therapy development.
