Abstract

Bacterial efflux pumps are one of the major mechanisms by which bacteria acquiring multidrug resistance. These pumps represent attractive therapeutic targets for compounds that are able to inhibit their function known by efflux pump inhibitors (EPIs). Recent work conducted by Bremner et. al. at the University of Wollongong has shown that the weak plant antimicrobial berberine administered by covalent linkage to an efflux pump inhibitor had improved its antibacterial efficacy.
In this project, the synthesis of a prototype dual-action antibacterial agent comprising the photosensitiser methylene blue covalently linked to a phenolic EPI from Dalea versicolor was attempted. The target molecule (Phen-EPI) has been designed for use in antimicrobial photodynamic therapy (APT).
In the course of this study, three compounds in the route of the overall synthesis were successfully synthesised and characterised. However due to the time constraints the overall aim to produce a prototype Phen-EPI has been not successfully yet attained.
