Abstract
Marsilea crenata Presl. is a typical plant in East Java, Indonesia which suspected contains estrogen-like substances. The present study aimed to investigate phytoestrogen stimulating activity of M. crenata leaves in increasing trabecular vertebrae bone density on osteoporosis female mice. Dexamethasone treatment (0,0029 mg/20gBW/day) was demostrated to facilitate the induction of osteoporosis. Mice were divided into five groups, each 6 mice: negative control, mice were given CMC Na 0,5% 0,4ml/20gBB/day; positive control, mice were given alendronate 0,026 mg/20gBW/day; treated groups, mice were given the n-hexane fraction of M. crenata on three different concentration, 1.54 mg/20gBW/day, 1.54 mg/20gBW/day combined with physical exercise, and 3.08 mg/20gBW/day. The result showed trabecular vertebrae thickness of each groups are 5.5 ± 0.7 μm (negative group), 8.2 ± 0.4 μm (positive group), 6.5 ± 0.4 μm (fraction of n-hexane at dose 1.54 mg/20gBW), 8.1 ± 0.6 μm (fraction of n-hexane at dose 1.54 mg/20gBW combined with physical exercise), and 8.0 ± 0.3 μm (fraction of nhexane at dose 3.08 mg/20gBW). Based on these results, the treatment of M. crenata n-hexane fraction have the activity in increasing trabecular vertebrae bone density on female mice, with the optimal dose 1.54 mg/20gBW combined with physical exercise and 3.08 mg/20gBW. This activity is probably due to phytoestrogens content in n-hexane fraction of M. crenata leaves, that can replace the function of estrogen in its bond with ER inside the bone.
