Abstract

Chlorella sp. is one of Allah creation that very usefull. Chlorella sp. contain steroid compound that can be used as anticancer and antioxidant. The objectives of this research is to know the toxicity level, antioxidant activity and to know the kind of the steroid compounds separation on ethyl acetate and petroleum eter fraction from Chlorella sp. microalgae isolated with Thin Layer Chromatography (TLC). Chlorella sp. was cultivated in Tauge Extract Medium (TEM) 4 % and harvesting of at stationary phase (10th day). Microalgae Chlorella sp. was extracted by maceration method using methanol solvent. The methanol extract was hydrolyzed with HCl 2 N and then partitioned with ethyl acetate and petroleum ether sovents. Ethyl acetate and petroleum ether fraction was tested by phytochemicals test to fine out the content of its activecompounds. The steroid compound fromethyl acetate and petroleum ether fraction was separated with preprarative TLC. The toxicity level of TLC isolates was determained by BSLT methods and antioxidant activity was determained by DPPH methods. The isolates was identified by FTIR spectrophotometer, UV-Vis spectrophotometer and LC-MS.

The result showed that TLC isolates of Chlorella sp. microalgae ethyl acetate and petroleum ether fraction was toxic againt Arthemia salina L. Brine shrimp larvae with LC50 value 14,9625 and 19,6889 ppm. Isolates of Chlorella sp. microalgae ethyl acetate and petroleum ether fraction has potential as anticancer or antitumor because its LC50 value under 30 pp. Isolates of Chlorella sp. microalgae ethyl acetate and petroleum ether fraction has antioxidant activity to DPPH wth EC50 value 77,78 and 73,82 ppm. Antioxidant activity of Chlorella sp. microalgae ethyl acetate and petroleum ether fraction was strong because its EC50 value under 100 ppm. Identified by FTIR spectrophotometer, UV-Vis spectrophotometer and LC-MS showed that of Chlorella sp. microalgae ethyl acetate and petroleum ether fraction was positive contain steroid compound.
